Single-Nucleotide Polymorphisms Related to Vitamin D Metabolism and Severity or Mortality of COVID-19: A Systematic Review and Meta-analysis

Systematic review and meta analysis of 12 studies examining associations between 31 single-nucleotide polymorphisms (SNPs) related to four vitamin D metabolism genes (VDR, GC, DHCR7, CYP2R1) and COVID-19 severity or mortality. Meta-analysis showed the VDR rs1544410 genotype and b allele were associated with increased odds of developing severe/critical COVID-19. Several VDR SNPs (rs731236, rs1544410, rs7975232, rs2228570) were associated with increased COVID-19 mortality.
18 meta analyses show significant improvements with vitamin D treatment for mortality1-13, mechanical ventilation1,6,7,12,14,15, ICU admission1,3,6,7,10,12,14-17, hospitalization5,12, severity2,4,6,11,18, and cases8,17,18.
Currently there are 122 vitamin D treatment for COVID-19 studies, showing 36% lower mortality [28‑43%], 19% lower ventilation [-3‑36%], 45% lower ICU admission [28‑58%], 19% lower hospitalization [9‑29%], and 17% fewer cases [9‑24%].
