In Vitro study showing effective inactivation of SARS-CoV-1 through multiple methods including UV light, heat, pH extremes, and chemical treatments in Vero E6 cells. Authors found that UVC light (254 nm) completely inactivated the virus after 15 minutes of exposure, while UVA light had no effect. Heat treatment at 75°C for 45 minutes achieved complete inactivation, while treatment at 56°C and 65°C required 90 minutes. Both formaldehyde and glutaraldehyde effectively inactivated the virus in a temperature-dependent manner, with glutaraldehyde showing complete inactivation after 1-2 days at room temperature or 37°C. Extreme pH conditions (pH ≤3 or ≥12) completely inactivated the virus. Authors also demonstrated that viral RNA alone or detergent-disrupted virions were not infectious.

Severe acute respiratory syndrome (SARS) is a life-threatening disease caused by a novel coronavirus termed SARS-CoV. Due to the severity of this disease, the World Health Organization (WHO) recommends that manipulation of active viral cultures of SARS-CoV be performed in containment laboratories at biosafety level 3 (BSL3). The virus was inactivated by ultraviolet light (UV) at 254 nm, heat treatment of 65 • C or greater, alkaline (pH > 12) or acidic (pH < 3) conditions, formalin and glutaraldehyde treatments. We describe the kinetics of these efficient viral inactivation methods, which will allow research with SARS-CoV containing materials, that are rendered non-infectious, to be conducted at reduced safety levels.
